Losing my job due s/o promotion

Location: Indiana. My spouse, (not married) and I work for the same company. We are both in salaried leadership positions. There is an opportunity for my spouse to move a level up in management which is would really be a great fit. If my so takes this position I will no longer be allowed to work for the company even on a different shift. I will not be "fired" but will be asked to resign is my assumption. There will be no severance package or pay to get me through until I find a new job. Will I get unemployment if I resign? Can I refuse to resign? I really dont know what to do, I have spent many years at this company and have a lot of sick pay and vacation accrued and I will just have to completely start over somewhere else.
